Calcitonin gene-related peptide regulates intracellular pH of cultured neuroblastoma cells. The function of calcitonin gene-related peptide (CGRP) in mammalian neuroendocrine cells is only beginning to be elucidated. In this study, the relationship between CGRP and intracellular pH (pHi) in cultured neuroblastoma cells was investigated using the fluorescent probe BCECF. Incubation of neuroblastoma N2A cells with CGRP (0.1-10 nM) caused a transient acidification in pHi by 0.1 units +/- 0.3 over 5 min, and the response was Ca(2+)-independent. The pH-sensitive dye 2′,7′-bis-(2-carboxyethyl)-5-(and 6)-carboxyfluorescein, which preferentially accumulates in the cytoplasm and thus represents a more cytosolic component of pHi, showed the same acidification induced by CGRP. Under the same conditions, in the presence of the CGRP antagonist hCGRP-(8-37), the response was not elicited. The effect of CGRP was inhibited by 30 microM Ni(2+), but not by 60 microM Ni(2+), suggesting that CGRP may activate a Ni(2+)-sensitive Ca(2+)-dependent Na+/H+ exchange. The change of pHi induced by CGRP was abolished by inhibition of N-type Ca2+ channels with either the antagonist omega-conotoxin GVIA (1 microM) or the specific antagonist omega-AgaIVA (30 microM). The pH response was not inhibited by an inhibitor of voltage-dependent Ca2+ channels, verapamil (10 microM). CGRP-mediated acidification was not affected by blocking the Cl- conductance with glibenclamide, and was not inhibited by intracellular application of the Cl- channel blocker niflumic acid (50 microM).
